薄皮甜瓜新品种京玉绿宝的选育

京玉绿宝是以白沙蜜5代自交系357为母本,以地方品种240提纯系选后代397为父本配制而成的薄皮甜瓜一代杂种。果实扁卵圆形,单果质量200~400g,果面光滑无棱,果皮深绿色,果肉白绿色,可溶性固形物含量11%~15%,早熟,抗逆性较强,适合保护地及少雨露地栽培,每667m2产量1600kg。已在北京、河北、辽宁、吉林、内蒙古等地示范推广3000hm2。     

Effect of hydrogen sulfide on myocardial fibrosis and PPARγ and NF-κB expression in rat model of diabetes

AIM: To explore the effects of hydrogen sulfide (H₂S) on the myocardial fibrosis in a rat model of diabetes and its mechanism.METHODS: Single intraperitoneal injection of streptozotocin (STZ) was utilized to establish a rat model of diabetes. Sodium hydrosulfide was used as an exogenous donor of hydrogen sulfide. Male SD rats were randomly divided into control group, STZ group, STZ+H₂S group and H₂S group. Eight weeks later, HE and VG staining methods were used to observe the collagen distribution and collagen volume fraction was measured by image analysis. The expression levels of type I collagen, PPARγ and NF-κB in the cardiac tissues were determined by Western blotting.RESULTS: Compared with control group, collagen distribution and the expression levels of type I collagen and NF-κB in the cardiac tissues were markedly increased (P<0.05), while PPARγ was significantly decreased in STZ group (P<0.05), but these indexes were reversed significantly in STZ+H₂S group (P<0.05). The expression levels of type I collagen, PPARγ and NF-κB had no significant difference between H₂S group and control group.CONCLUSION: Hydrogen sulfide attenuates cardiac fibrosis in diabetic rats, and its mechanism may be related to PPARγ-NF-κB signaling pathway.     

基于改进GM(1,N)模型的我国大豆价格影响因素分析及预测研究

大豆是我国重要的粮食作物和油料作物,其价格对于国民经济尤其是农业经济的影响意义深远。大豆价格的稳定对于我国大豆市场的健康发展有着重要的现实意义。在灰色理论的基础上,提出了一种改进GM(1,N)大豆价格预测模型,首先运用灰色关联分析法对我国大豆价格的影响因素进行分析,选择主要的影响因素;再将这些影响因素作为模型的相关因素变量,构建GM(1,N)大豆价格预测模型。采用2010-2015年的大豆数据进行实证研究,模型选取国内大豆自给量、世界大豆产量、国民消费价格指数、消费者信心指数4个变量作为相关因素变量;模型预测误差为2.10%,预测精度较高,能够较好地掌握大豆价格的变化规律,可以为大豆价格市场预测及国家宏观政策的制定提供理论指导。     

Mycoplasma pneumoniae induces IL-1β production through activating NLRP3 inflammasome by ROS in RAW264.7 cells

AIM: To investigate whether Mycoplasma pneumoniae (Mp)-induced interleukin-1β (IL-1β) production in RAW264.7 cells is through the activation of NLRP3 inflammasome via reactive oxygen species (ROS). ME-THODS: RAW264.7 cells were randomly divided into 3 groups. In normal group, RAW264.7 cells were treated without Mp. In model group, RAW264.7 cells were treated with 1∶ 10 multiplicity of infection (MOI) of Mp. In NAC group, RAW264.7 cells were pretreated with N- acetylcysteine (NAC) at a concentration of 5 mmol/L for 30 min before infection with Mp. The RAW264.7cells were infected with Mp (1∶ 10 MOI) for 4, 8, 16 and 24 h in model group and NAC group, respectively. The intracellular ROS level was analyzed by flow cytometry. The mRNA expressions of NLRP3, ASC and caspase-1 were detected by real-time PCR. The protein levels of NLRP3, ASC and caspase-1 p20 were determined by Western blot. The levels of pro-inflammatory cytokine IL-1β in the supernatant were measured by ELISA. RESULTS: Compared with normal group, the production of ROS were significantly increased at 4, 8, 16 and 24 h after infection, the mRNA expression of NLRP3, ASC and caspase-1 were increased at 8, 16 and 24 h after infection, the protein levels of NLRP3, ASC and caspase-1 p20 were increased at 16 and 24 h after infection, and the releases of IL-1β were increased at 24 h after infection in model group (P<0.01). Compared with the model group, the level of ROS in NAC group decreased, so as the expression of NLRP3, ASC and caspase-1 at mRNA and protein levels and the releases of IL-1β in the supernatant at the corresponding time points. CONCLUSION: Mp may stimulate the ROS production to activate NLRP3 inflammasome in RAW264.7 cells.     

浅议慕课及其对精品课程建设的启示

慕课(MOOC)在学习成本低、学习效率高、交互性强等方面受到了广大学习者的青睐,逐渐成为终身学习的一种渠道。精品课程建设是高校教育与教学改革工程必不可少的环节,精品课程建设第二期工程——精品资源共享课正在建设过程中。慕课运行方式对我国精品资源共享课建设具有一定借鉴意义。     

龙眼DCL4的亚细胞定位分析及其对外源ABA和SA的响应

为了研究DlDCL家族中DlDCL4基因在龙眼中的功能,本研究通过氨基酸序列比对、系统发育进化树的构建、亚细胞定位验证、实时荧光定量PCR（qPCR）等方法,对其进行初步探究。结果表明：遗传进化分析显示,DCL4氨基酸序列在所选的物种间相似度高于50%,其中龙眼与甜橙（Citrus sinensis）的亲缘关系最近。在激光共聚焦显微镜观察下,洋葱细胞核发出绿色荧光,其他部位不发出荧光,表明龙眼DCL4蛋白定位于细胞核中。在24 h内,100 μmol/L的外源脱落酸（ABA）对 DlDCL4都有极显著的下调效果;100 μmol/L的水杨酸（SA）对其表达量有显著的上调效果,其中8 h时表达量最大,提示 DlDCL4对不同激素的响应效果不同且不同处理时长下响应效果也会有影响。     

百香果番茄复合果酒的工艺研究

以百香果和番茄为主要原料,对百香果番茄复合果酒的加工工艺进行了研究。结果表明,最佳的工艺参数为：百香果番茄汁液(V:V)1:2、初始糖度23.0 °Bx、酵母添加量0.25%、发酵温度22 ℃。这四种因素对百香果番茄复合果酒影响的主次顺序为百香果番茄汁V:V >酵母添加量>发酵温度>初始糖度。百香果番茄复合果酒的成品指标为：酒度10.4 °(V%),残糖4.9%,pH值为4.3,呈柠檬黄色,清澈透明无悬浮物、果香酒香浓郁、丰满爽口有新鲜感。     

微生物对鱼粉中组胺含量的影响研究

我国鱼粉产品质量参差不齐,由于原料问题组胺含量普遍高于国际先进水平,过高的鱼粉含量易造成食品安全问题。为了降低过高的组胺含量对鱼粉品质的影响,本实验通过使用微生物参与降解鱼粉产品中的组胺。实验将降解时间与微生物添加比例作为变量,测试出较为适宜微生物降级组胺的条件,以及该条件下的实际效果。实验结果表明在设置的6个时间中(以6小时为时间梯),24h为较适宜的降解时间；在较适宜的降解时间中(24h)较适宜的微生物添加比例为10:1。此时鱼粉组胺含量为1622mg/kg,相较未加入微生物处理的鱼粉组胺含量(5298 mg/kg)降低69%。实验结果证明了微生物处理鱼粉中组胺具有显著的效果,为鱼粉生产、储存技术的改良提供了参考依据。     

利用正交实验设计研究磺胺甲恶唑在渔业水体中的消解动态规律

为了探究磺胺甲恶唑(sulfamethoxazole,SMZ)在渔业水体中的自然降解规律,本试验选取了养殖水环境中常见的因素(温度、pH和敞蔽条件),通过正交实验探究这三个因素对SMZ在自然养殖水体中消解动态的影响。试验初期各处理SMZ浓度保持一致,实验周期内定期取水样检测水体中SMZ的浓度变化,符合一级反应动力学方程,计算各处理中SMZ药物的半衰期和消除率。结果表明,18个处理组SMZ的半衰期及其消除率变化范围为4.88-12.89d,28%-77%。方差分析得出：在实验水平范围内,温度、pH因素对SMZ的消解结果有显著影响,但敞蔽的影响较小,因此提高养殖水体高温度、调节酸碱性pH等条件均有利于促进SMZ的降解。
